How to reconstitute a peptide: a step-by-step guide
Peptides are supplied in lyophilized form — a dry powder obtained by freeze-drying. Before any handling in a research context, this powder must be brought back into solution: this is reconstitution. Done well, it determines the accuracy of every draw and the stability of the product. Here is the method, step by step.
What reconstitution is for
Freeze-drying removes water to keep the peptide stable over time. To handle it, you redissolve it in a suitable solvent — most often bacteriostatic water. The goal is a known, homogeneous concentration, expressed in mg/ml (or mcg/ml), so you can draw exact volumes.
Materials
- The vial of lyophilized peptide (sealed).
- Bacteriostatic water (sterile water with 0.9% benzyl alcohol, which limits bacterial growth over several days).
- A sterile syringe (often an insulin syringe graduated in U-100 units).
- An alcohol swab to disinfect the stopper before each draw.
The steps
- Let everything reach room temperature. A peptide taken from the cold handles better and limits condensation.
- Disinfect the stoppers of both the peptide and the bacteriostatic water with an alcohol swab.
- Draw the chosen volume of water with the syringe (see the calculation below).
- Inject the water slowly against the vial wall, never straight onto the powder: peptides are fragile, and a harsh jet can degrade them. Let it run gently.
- Do not shake. Gently swirl the vial (circular motion) until fully dissolved. The solution should turn clear.
A solution that stays cloudy, colored or contains particles after full dissolution is a signal not to ignore.
Calculating the concentration
The rule is simple:
Concentration = amount of peptide (mg) ÷ volume of water added (ml).
For example, 10 mg reconstituted in 2 ml gives 5 mg/ml (i.e. 5000 mcg/ml). The more water you add, the more diluted the solution — and the larger the volume to draw for the same amount, which improves reading precision on the syringe.

Storage after reconstitution
Once reconstituted, keep the peptide in the refrigerator (2–8 °C), away from light. Reconstituted with bacteriostatic water, it generally keeps for several weeks. The un-reconstituted powder keeps far longer — ideally in the freezer. Avoid repeated freeze–thaw cycles.
Common mistakes
- Shaking the vial: foam and possible degradation. Swirl, don't shake.
- Injecting the water too fast onto the powder.
- Forgetting to disinfect the stopper before each draw.
- Choosing too little water: the solution becomes very concentrated and the volumes to draw become tiny, therefore imprecise.
